常用生化试剂作用:
1、蛋白酶K:能水解消化蛋白质,特别是与DNA结合的组蛋白,在尿素和SDS中稳定。一般工作浓度是50—100μg/ml,推荐反应缓冲液:50mM Tris-HCl (pH7.5),10mM CaCl2。
2、SDS:十二烷基硫酸钠,溶解细胞膜上的脂类与蛋白质,因而溶解膜蛋白而破坏细胞膜,并解离细胞中的核蛋白,SDS 还能与蛋白质结合而沉淀。
3、IPTG:异丙基-β-D-硫代半乳糖苷, 常用于蓝白斑筛选及IPTG诱导的细菌内的蛋白表达等。IPTG和乳糖的结构相似,所以它和乳糖一样,可以与乳糖操纵元的阻遏蛋白结合,
使阻遏蛋白的空间构像变化,四聚体解聚成单体,失去与操纵子特异性结合的能力,从而解除了阻遏蛋白的作用,使其后的基因得以转录合成利用乳糖的酶类。与乳糖不同的是,
IPTG不被 β-半乳糖苷酶水解。常与X-GAL一起用于蓝白斑筛选。作用*的诱导剂,不被细菌代谢而十分稳定。
4、X-gal:5-溴-4-氯-3-吲哚-β-D-半乳糖苷,是IPTG的显色试剂,在IPTG的催化下显蓝色。常跟IPTG一起用与蓝白斑筛选。
5、G418:一种抗生素,对几乎所有的细胞都有毒性,是稳定转染zui常用的选择试剂。当neo基因被整合进真核细胞基因组合适的地方
6、DMSO:二甲基亚砜。实验室常用于作液体层析溶剂,同时用作测试物质紫外消光值上时用作参照物。能溶于所有烷烃和烯烃。
7、曲拉通X-100:TritonX-100,用的细胞裂解液成分之一,在保护蛋白活性方面有一定作用, 能力介于NP40和SDS之间,偏向于NP40。
8、NP-40:很温和的去垢剂,1%浓度的基本可以破坏掉胞膜,而对核膜破坏的作用弱,结合特定的buffer可以获得胞浆蛋白。
9、BSA:牛血清白蛋白, 主要起维持渗透压作用、PH缓冲作用、载体作用和营养作用。在动物细胞无血清培养中,添加白蛋白可起到生理和机械保护作用和载体作用。常用于蛋白定量中的内参和抗体稀释液。
10、甲酰胺: 甲酰胺被用作凝胶电泳中RNA的稳定剂,也用于稳定毛细管电泳中的变性单股DNA。
