    HPLC色谱柱保养和储存

     

    HPLC柱具有有限的寿命。建议您使用标准定期监测色谱柱的保留特性和性能。如果您打算长时间储存​​色谱柱,请使用抑制微生物生长的流动相(含有sodium azide或高浓度甲醇,乙腈等的流动相)。为获得性能和色谱柱寿命,请遵循以下建议。

    • 定期监控色谱柱的性能。
    • 仅在可混溶的流动相之间切换。
    • 避免在色谱柱中沉淀盐。
    • 使用过滤和脱气的流动相。
    • 不要让色谱柱变干。不使用时请将其盖紧。
    • 对于长期储存,使用抑制细菌和霉菌生长的流动相。
    • 按标签上指示的方向使用该列。异常高的操作压力可能表明插入的入口玻璃料堵塞,并且可以通过使流过柱的流量反转10-20mL来清除。
    • 使用Hamilton保护柱去除可能终身性结合分析柱的颗粒物质或杂质。

    恢复色谱柱性能

    Flush with 50 mL 0.1 N sodium hydroxide. Equilibrate with starting phase for ~ 200 column volumes.

    RCX-30

    Flush with 150 mL 0.1 N sodium hydroxide. Equilibrate with starting phase for ~ 200 column volumes.

    HC-40

    Ca2+  Flush with 1% calcium chloride at 0.1 mL/min overnight. Equilibrate with starting phase for ~ 200 column volumes.

    HC-75
