原理

TF Luciferase Reporter Vectors

pTF-Luc报告载体是一系列基于萤火虫荧光素酶的报道构建体,用于定量测量细胞中转录因子(TF)的活性。每个载体含有顺式元件(DNA结合序列),小启动子和萤火虫荧光素酶基因。当TF被激活时,它与顺式元件结合并导致荧光素酶基因表达的诱导。因此,荧光素酶活性代表TF的活性。

类别: 氧化应激 

活性氧(ROS)在代谢和生理过程中产生,并且有害的氧化反应可能发生在通过酶和非酶抗氧化机制去除它们的生物体中。在某些条件下,不能防止氧化剂的增加和抗氧化剂的减少,并且发展氧化或超过100种疾病。

 

抗氧化剂分子可预防或抑制这些有害反应。不同抗氧化剂的血清(或血浆)浓度可以在实验室中单独测量,但是测量是耗时的,劳动密集的,昂贵的并且需要复杂的技术。由于不同抗氧化剂分子的测量分别不实用且其抗氧化效果是相加的,因此测量样品的总抗氧化能力,这称为总抗氧化能力(TAC),总抗氧化活性(TAA),总抗氧化能力(TAOP),总抗氧化状态(TAS),总抗氧化反应或其他同义词。

在测试中,样品中的抗氧化剂将深蓝绿色的ABTS基团减少为无色减少的ABTS形式。660nm处吸光度的变化与样品的总抗氧化剂水平有关。该试验用稳定的抗氧化标准溶液校准,该标准溶液传统上称为Trolox Equivalent,它是维生素E类似物。
