BioChain的cfPure®无细胞DNA提取试剂盒专为快速有效纯化无循环细胞DNA（cfDNA）而设计。该系统利用二氧化硅涂层的顺磁颗粒来纯化无细胞DNA，从小于1 mL到大于10 mL的血清或血浆。开发缓冲液以确保有效回收100bp-500bp的DNA片段，以大限度地回收无细胞DNA。回收的DNA适用于广泛的下游应用，例如亚硫酸氢盐测序，NGS和qPCR。

基于磁珠的快速协议

cfPure®磁珠净化的优点

与用于分离cfDNA的其他技术相比，磁珠提供许多益处。珠子比玻璃纤维过滤器更有效地结合DNA，从而产生更高和更一致的产量。另外，由于不使用过滤器和真空歧管，因此在提取过程中不存在细胞颗粒堵塞这些物品的风险。这种堵塞问题特别涉及富含蛋白质的大体积样品，例如cfDNA研究中常用的血浆。

cfDNA由终用户通过NGS验证

cfPure®fCRDNA提取试剂盒提取的cfDNA非常适用于NGS等下游应用。在第三方研究中，使用BioChain的cfPure®（B），竞争对手T（T）和竞争对手Q（Q，基于柱）的cfDNA提取试剂盒从4个供体的10ml人血浆中提取cfDNA。然后为每个样品构建文库，并进行全基因组测序。从数据分析来看，三种提取方法并未显示出显着的性能差异：插入大小（图3），定位率，全基因组覆盖率，替代率和GC偏差（图4）。

cfDNA的可重复和有效回收

我们优化的基于磁珠的协议使用户能够扩展协议，适用于100μl至10 ml的任何样品体积，具有高度可重复的结果和有效的cfDNA回收率（表1和2）。

自动化友好

BioChain 的 PCR Ready First Strand cDNA 可从不同的成人正常组织中获得。每个 PCR 反应 1 µl cDNA 就足够了。

产品描述

第一链 cDNA 是从从各种已证实的成人正常组织、人类患病和肿瘤组织、小鼠、大鼠、猴和植物组织中分离出来的 RNA 合成的。用于 cDNA 合成的总 RNA 是通过改进的硫氰酸胍技术分离的。10 µg 总 RNA 通过 oligo dT 引物和 MMLV 逆转录酶进行逆转录 (RT)，最终体积为 40 µl。通过在 65°C 加热 10 分钟停止 RT 反应。cDNA 在 1x RT 缓冲液（1x RT 缓冲液：50 mM Tris-Cl，pH 8.3，75 mM KCl，3 mM MgCl2，10 mM DTT）中递送。1 µl cDNA 足以进行一次 PCR 反应。人类网格蛋白 cDNA 的 5' 末端（一个 6 kb 基因）已通过 PCR 从所有这些 cDNA 中扩增。
