质量控制分析
单位表征测定
单位活性使用2倍系列稀释法测量。在1X反应缓冲液中制备酶的稀释液并加入到20μL含有双链DNA的片段和1X反应缓冲液的反应中。将反应在25℃(室温)孵育30分钟,在冰上浸入,并在用溴化乙锭染色的1%琼脂糖凝胶上分析。
蛋白质浓度(OD 280)测量使用使用2.0mg / ml BSA样品(Pierce Cat#23209)标准化的Nanodrop ND-2000分光光度计
在OD 280处分析2.0μL酶样品,并用产品储存溶液冲切。使用34,280的消光系数和73,606道尔顿的分子量将观察到的3个重复样品的平均测量结果转换为mg / mL。
SDS-Page(物理纯度评估)
将2.0μL酶溶液加载到变性的4-20%Tris-甘氨酸SDS-PAGE凝胶上,该凝胶侧翼为宽范围的MW标记和2.0μL的1:100稀释的样品。电泳后,将凝胶染色并比较样品以确定物理纯度。该测试的验收标准要求浓缩样品中污染带的总量不超过稀释样品中目的蛋白质条带的质量,确认浓缩样品的纯度大于99%。
污染测试
单链核酸外切酶活性
将含有10,000cpm放射性标记的单链DNA底物和10μl酶溶液的50μL反应物在37℃下温育4小时,导致TCA可溶性计数的释放小于2.0%。
双链核酸外切酶活性将含有5,000cpm放射性标记的双链DNA底物10μl的50μL反应液和在37℃下孵育4小时的酶溶液导致少于1.0%的TCA可溶性计数释放。
双链核酸内切酶活性将含有0.5μgpBR322 DNA和100单位酶溶液的50μL反应液在37℃下温育4小时,结果通过琼脂糖凝胶电泳测定,没有视觉上可辨别的转化为带切口的环状DNA。
大肠杆菌 16S rDNA污染测试
将酶溶液的重复样品变性,并使用对应于16S rRNA基因座的寡核苷酸引物,在TaqMan qPCR测定中筛选污染性大肠杆菌基因组DNA的存在。测试的验收标准是由3个重复的无模板对照样品的平均值产生的阈值循环计数(C t)。基于无模板对照 Ct值与标准曲线数据之间的相关性,该测定的检测限为<10拷贝基因组/样品。
