OptiGene 的等温预混液提供 DNA 扩增速度

OptiGene 的等温预混液（ISO-001 和 ISO-004）与 NEB 的 WarmStart LAMP 试剂盒（DNA 和 RNA）进行了比较

 

从人类 gDNA (Sigma) 扩增 DNA 靶标 (VKORC1)。25μl LAMP 反应在 65ºC 下在实时热循环仪 (Roche LC-480) 上进行 60 分钟，一式两份。“产生结果的时间”定义为扩增荧光值达到最大荧光 10% 的阈值的时间。

使用 LAMPDesigner® 设计了靶向 VKORC1 基因的 LAMP 引物。

OptiGene 的等温预混液提供RNA 扩增速度

OptiGene 的等温预混液（ISO-001 和 ISO-004）与 NEB 的 WarmStart LAMP 试剂盒（DNA 和 RNA）[Lot:0021609] 进行了比较。ISO-001 和 ISO-004 不含特定的逆转录酶 (RT)。为了与 NEB 混合物（包含单独和特定的逆转录酶）进行真正的比较，将 0.5U AMV-RT 添加到 OptiGene Master Mix 反应（ISO-001 inc. RT 和 ISO-004 inc. RT）中。还比较了标准 ISO-001 和 ISO-004 RT-LAMP 反应，以显示混合物具有优异的固有逆转录酶活性。
