Description

pLVX-IRES-ZsGreen1 is an HIV-1-based, lentiviral expression vector that allows the simultaneous 

expression of your protein of interest and ZsGreen1 in virtually any mammalian cell type, 

including primary cells. ZsGreen1 is a human codon-optimized variant of the reef coral 

Zoanthus sp. green fluorescent protein, ZsGreen. The vector expresses the two proteins from 

a bicistronic mRNA transcript, allowing ZsGreen1 to be used as an indicator of transduction 

efficiency and a marker for selection by flow cytometry.

Expression of the bicistronic transcript is driven by the constitutively active human 

cytomegalovirus immediate early promoter (P

CMV IE) located just upstream of the MCS. An 

encephalomyocarditis virus (EMCV) internal ribosome entry site (IRES), positioned between 

the MCS and ZsGreen1, facilitates cap-independent translation of ZsGreen1 from an internal 

start site at the IRES/ZsGreen1 junction (1)

Description

pLVX-AcGFP1-N1 is an HIV-1-based, lentiviral expression vector that allows you to express 

your gene of interest fused to AcGFP1, a green fl uorescent protein derived from Aequorea 

coerulescens. Genes cloned into the multiple cloning site (MCS), located upstream of the 

AcGFP1 coding sequence, are expressed as N-terminal fusions of the AcGFP1 protein. 

Expression of the fusion protein is driven by the constitutively active human cytomegalovirus

immediate early promoter (P

CMV IE) located just upstream of the MCS. Lentiviral particles 

derived from the vector allow the expression of AcGFP1 fusion proteins in virtually any cell 

type, including primary cells. The unmodifi ed vector expresses AcGFP1, and may be used to 

produce marker virus to optimize infection protocols.

The pLVX-shRNA1 Vector is a lentiviral expression vector that can be used to generate high titers of lentivirus for transducing dividing or nondividing mammalian cells. Transduced target cells will express a user-generated shRNA from the U6 promoter for the purpose of RNAi-mediated gene silencing. pLVX-shRNA1 Vector can be packaged into high-titer, replication-incompetent, VSV-G-pseudotyped lentivirus upon transfection into 293T cells, using the Lenti-X™ HTX Packaging System. The resulting lentivirus can then be used to transduce virtually any mammalian cell type.
