Exonuclease T
- Also known as RNase T
- Single-stranded (ssDNA or RNA) specific exonuclease
- Catalyzes the removal of nucleotides from linear single-stranded DNA or RNA in the 3′ to 5′ direction
Exonuclease T can be used for:
- Removal of 3′ overhangs of double-stranded DNA to generate blunt-ends*
- Removal of single-stranded primers in PCR reactions prior to DNA sequencing or SNP analysis
- Removal of single-stranded primers for nested PCR reactions
- Removal of single-stranded DNA, leaving behind double-stranded DNA in the sample

Exonuclease III (E. coli)
- Double-stranded DNA specific exonuclease
- Initiates at the 3′ termini of linear double-stranded DNA with 5′ overhangs or blunt ends and 3′ overhangs containing less than four bases
- Initiates at nicked sites in double-stranded DNA
- Catalyzes the removal of nucleotides from linear or nicked double-stranded DNA in the 3′ to 5′ direction
Exonuclease III is ideal for:
- Site-directed mutagenesis
- Preparation of single-stranded DNA for dideoxy sequencing
- Preparation of nested deletions in double-stranded DNA

This product replaces T5 Exonuclease (NEB #M0363)
- Double-stranded DNA specific exonuclease and single-stranded DNA endonuclease
- Initiates at the 5′ termini of linear or nicked double-stranded DNA
- Cleaves linear or nicked double-stranded DNA in the 5′ to 3′ direction
T5 Exonuclease is ideal for:
- Removal of incomplete ligation products from ligated circular double-stranded DNA
- Degradation of denatured DNA from alkaline-based plasmid purification methods for improved DNA cloning
- Degradation of contaminating linear and nicked DNA in plasmid samples
