Thermostable USER (Uracil-Specific Excision Reagent) III Enzyme generates a single nucleotide gap at the location of a uracil residue. It is active between 50-75°C (optimal activity observed at 65°C).

Thermostable RNase H
Thermostable RNase H specifically recognizes and cleaves the phosphodiester bonds of an RNA strand in an RNA-DNA hybrid while leaving the DNA strand intact. This thermostable nuclease exhibits the same enzymatic properties as E. coli RNase H, but is active at much higher temperatures.
Recombinant enzyme supplied with 10X Reaction Buffer
Does not digest single or double-stranded DNA
Active and stable above 37°C over a broad temperature range, with activity increasing with temperature
Requires magnesium chloride for activity
Inactivated with proteinase K treatment or addition of excess EDTA

Thermostable DNA and RNA endonuclease
Catalyzes the cleavage of 5′ DNA and RNA flaps from branched double stranded DNA substrates, creating a 5′ phosphate terminus
FEN 1 products can be ligated by DNA ligase to create double stranded DNA

Thermostable oxoguanine glycosylase
Bifunctional DNA glycosylase with DNA N-glycosylase and AP lyase activities
N-glycosylase activity releases 8-oxo-7,8-dihydroguanine (8-oxoG), generating an AP site. The AP-lyase activity cleaves 3′ to the AP site leaving a 5′ phosphate and a 3′-phospho-α, β-unsaturated aldehyde.
